To test tissue distribution, we injected CD 1 mice intravenously with either radiolabeled trastuzumab (125I-trastuzumab) or 125I-H43 at an equivalent dose of 10 mg/kg. Blood samples were collected 30 minutes, 1 hour, 4 hours, 7 hours, 24 hours, 48 hours, and 72 hours later. The mice were then sacrificed and perfused with cold phosphate-buffered saline for ~ 8 minutes. Various tissues were harvested and radioactivity was measured to assess tissue distribution. The results are shown in Figure 7. We found that, after 30 minutes, "the distribution of H43 in the brain was 3.5-fold greater than that of trastuzumab"; after 60 minutes, the distribution was 2.5-fold greater; and after 72 hours, the distribution was 2.3-fold higher.
